Sometimes, yes. It is easy to see with some people what desktop OS 
they're using, such as:

X-Mailer: Apple Mail (2.552)
X-Mailer: Microsoft Outlook Express 6.00.2800.1106
User-Agent: Mozilla/5.0 (X11; U; Linux i686; en-US; rv:1.2.1)

But for the people hiding behind some type of web based email, mutt or 
pine you really can't tell what they're using as their desktop OS unless 
you ask and they answer honestly.
